Research Assistant/ Officer: Research Team – Cartherics Pty Ltd
Proposed start date 4 January 2022.
The successful applicant will have at least a postgraduate degree such as a B.Sc Hons and/or PhD in Biomedical Science or equivalent and ideally 3 or more years of appropriate laboratory experience.
TBD depending on qualifications and experience, range $78,000 - $102,000.
Awarded on annual Performance Review.
The goal of the Cartherics Research Team is to develop a pipeline of important novel functional enhancements for the Company’s immune effector cells, particularly for the more advanced iNK program, but also to improve the function and safety profile of its iT cells. The Research Team is also exploring the induction from KI/KO gene-edited iPSC, of effector cells of the innate immune system such as macrophages and eosinophils. Both the adaptive and innate immune systems synergize remarkably in the defence against infection – a logical transition is to harness both for cancer immunotherapy.
There are already some well-developed programs within the Research Team. The new appointment is critical for not only taking on new studies but indeed for the successful continuation of the current projects which include:
- Creating novel signal transduction “CAR” constructs, tailored to be more effective in NK cells or T cells - “one size does not fit all”
- Development of dual CAR’s
- Improving iNK function by deletion of genes to increase cytotoxicity function
- Development of CAR-iT cells from iPSC
- Reactivation of endogenous thymus production of naive T cells using progenitor cells derived from iPSC
- Induction of innate immune cells (macrophages)
The primary requirement is a Research Assistant to provide much needed technical support for these projects. Not only are they supporting the projects within the Research Team, but they and other Team members provide great input into other Cartherics programs, including iNK differentiation from gene-edited iPSC, trouble-shooting scale-up method of iNK and CAR-T cell manufacture, molecular biology skills, CRISPR/Cas9 gene editing, flow cytometry etc. The current resources are clearly at beyond maximum capacity.
The successful applicant will work closely with all members of the Research Team, in particular with the deputy Lab Manager, and have the appropriate knowledge base and “hands-on” skills in one or more of the following technologies:
- Isolation, activation, in vitro expansion and functional analysis of human immune effector cells, including use of multi-parameter flow cytometry and in vitro cytotoxicity assays
- Experience in the in vitro culture, maintenance and directed differentiation of iPSC, ideally with an understanding of NK and/or T cell biology
- General molecular biology methods including the production and use of CRISPR Cas9 gene editing and associated cell biology; PCR
- Experience in animal handling (mice), particularly in cell infusion (i.v, i.p, subcutaneous); monitoring of cancer growth
- Organisational skills required for data management and documentation of standard operating procedures
- They will relish working in a challenging dynamic, interactive and very exciting environment.
The initial specialized technologies to be upskilled, under guidance, by the successful applicant will be:
- flow cytometric analysis of iNK cells
- in vitro xCELLigence cytotoxicity
- culture, cloning and iNK differentiation of gene-edited iPSC
- invitro cytokine analysis (Luminex microarray).
These are critical core technologies for the Cartherics Research Team.
Reports to Chief Scientific Officer (CSO) and Deputy Scientific Officer (DSO).
